Optimization of hydrolysis conditions for xylans and straw hydrolysates by HPLC analysis

Abstract Oligosaccharide analysis is commonly done by acid hydrolysis and following HPLC analysis. A major problem the incomplete of oligosaccharides disaccharides increasing formation volatile furfural from pentose monomers hydroxymethylfurfural (HMF) hexose monomers. This paper optimizes conditions approaches proposes a method for oligosaccharide quantification. The optimal condition model xylan corn cob was found to be 100 °C temperature, 120 min time, 2 wt% sulfuric concentration. Under these conditions, total free bound xylose yield 77.4% hemicellulose conversion 87.4% respectively; no degradation products were found. beech wood 65.1% 70.5% For pentosan hydrolysate, further optimized (110 °C, 60 min, H SO 4 ). Standard addition hydrolysation showed similar rates (< 2% deviation); matrix effects detected.
